Method Description

This is an enzymatic method based on the reduction of oxalate by oxalate oxidase. The reaction releases hydrogen peroxide, which in the presence of peroxidase reacts with a dye to give a colored end point that is measured using a BioTek EPOCH plate spectrophotometer at 590 nm. Patient Preparation

Fasting (12 hours). Patient should avoid taking vitamin C supplements for 24 hours prior to collection.

Collection & Handling

Collection Instructions

Place specimen on wet ice immediately.
Centrifuge for 10 minutes at 3,500 rpm at 4°C within 1 hour of collection and aliquot plasma into a plastic vial.
Adjust the pH of the plasma specimen to a pH of 2.3-2.7 with approximately 10 µL concentrated (12M) hydrochloric acid (or 20 µL of 6M HCl) per 1 mL plasma.

Handling Information

Store and send frozen.

Additional Information

Nonacidified specimens can be accepted if the heparinized plasma is properly frozen. However, a disclaimer will be added in nonacidified plasma: Sample was received nonacidified and frozen. In nonacidified samples oxalate values may increase spontaneously.
